NHIBITORY EFFECT OF PIPERONYLIC ACID ON CHOLINESTERASE’S IN VITRO AND IN SILICO

The present study investigates the inhibitory effect of piperonylic acid on cholinesterases through an in vitro and in silico analysis. Cholinesterases, including acetylcholinesterase (AChE) and butyrylcholinesterase (BChE), play crucial roles in the regulation of cholinergic neurotransmission. Dysregulation of these enzymes has been linked to neurodegenerative disorders such as Alzheimer's disease. Piperonylic acid, a naturally occurring compound, has shown promising bioactive properties, making it a potential candidate for therapeutic interventions targeting cholinesterase activity. In this research, an in vitro assay was conducted to measure the inhibitory activity of piperonylic acid on AChE and BChE enzymes. Additionally, molecular docking and computational simulations were performed to gain insights into the binding interactions between piperonylic acid and the active sites of cholinesterases. The results revealed that piperonylic acid exhibited significant inhibitory activity against both AChE and BChE in a dose-dependent manner. Furthermore, the in silico analysis suggested that piperonylic acid forms stable interactions with key residues within the active sites of the enzymes, providing a molecular basis for its inhibitory effects. These findings highlight the potential of piperonylic acid as a lead compound for the development of novel cholinesterase inhibitors for therapeutic applications in neurodegenerative diseases.
